Chymotrypsin
digestive enzyme found in pancreatic juice of the duodenum, catalysing proteolysis; cleaves proteins on the N-terminal of large aliphatic amino acids (tyrosine, tryptophan, phenylalanine)
Elastase
serine protease that breaks down elastin
Catalytic triad
a set of three coordinated amino acids found within the active site of some enzymes; can be conserved across classes of enzymes (including spatial conservation)
Sequence identity
proportion of amino acids that are identical between two protein sequences
Sequence similarity
a measure of how closely related two amino acid sequences are, by class of amino acid (hydrophobic, hydrophilic, ...)
TEV protease
highly sequence-specific protease used for the controlled cleavage of fusion proteins
thrombin
blood coagulation factor and sequence-specific serine protease; less specific than TEV, and can cleave almost any protein if incubated for long enough
Affinity chromatography
a technique to separate biomolecules through the selective binding of a molecule, and then eluting the column to release the molecule of interest; the process used in the practical session
Chymotrypsinogen
an inactive form of chymotrypsin, which gets cleaved to produce the active form
Trypsinogen
precursor form of trypsin, produced in the pancreas
Proelastase
inactive precursor form of elastase, produced by the acinar cells of the pancreas
Subtilisin
a serine protease obtained from Bacillus subtilis
Bortezomib
anti-cancer drug inhibiting the 26S proteasome, with high specificity and affinity; the boron atom transitions between trigonal and tetrahedral geometries
